VariOr® Gen

determination of genetic predispositions to periodontitis

Periodontitis is an inflammatory disease caused by one or more bacterial pathogens. Thanks to our VariOr® Dento test, we are able to identify the bacteria responsible for the disease. Subsequently, based on the knowledge of the amount and proportion of bacteria, appropriate and targeted treatment can be used. The success of the therapy can be checked by follow-up tests of VariOr® Dento.

In addition to microbial causes, however, there are other factors that affect the likelihood of periodontitis and its course. Some of these factors are hereditary. These include, for example, the mode of the patient's immune response - inflammation and the local and systemic immune response. The effectiveness of the immune response has a demonstrable effect on the extent of periodontal destruction and thus on the course of the disease. Inherited predispositions to periodontitis can be determined using the VariOr® Gen test.

What is a test for genetic predispositions to periodontitis?

Using the test, it is possible to determine the degree of genetic predisposition to periodontitis and especially to predict its course. The assay detects risk alleles of genes encoding the proinflammatory cytokine interleukin 1 (IL-1 alpha allele 2, IL-1 beta allele 2) and a specific serotype of the major histocompatibility system HLA-DR4.

What the test of genetic predispositions brings

The test provides information on the patient's hereditary predisposition to periodontal disease. Based on this information the doctor can individually adjust the care provided to the patient, the frequency of check-ups, etc. The patient, who is aware of the high personal risk of the disease, then approaches the preventive or therapeutic measures proposed by the doctor more willingly. Due to the high frequency of non-acceptance of dental implants in patients (especially smokers) with the risk allele IL-1beta, the use of the test is also indicated before more demanding surgical procedures.

Who is the test for?

The test is intended both for general dentists, including pediatricians, for early detection of patients at high risk of periodontitis (recommended as part of the initial examination), and for periodontists, to detect all causes that contribute to the patient's current periodontal condition. The doctor can individually adjust the care provided to the patient, recommend preventive measures, set an increased frequency of inspections, etc. In patients with periodontitis, the test can be performed with the same sample as VariOr® Dento. In other patients, only a buccal mucosa swab is sufficient.

Of course, the test can also be performed at the request of the patient, for example in children of parents with periodontitis it is possible to confirm or refute the transmission of genetic predispositions to the child and, if necessary, take the measures to prevent periodontitis or limit the consequences of the disease.

  • implantologists before surgery (determining the risk of non-admission)

Main advantages of the test:

  • the test is performed from the same sample as VariOr® Dento - the patient is therefore not burdened by double sampling; in the case of patients without clinical manifestations of periodontitis, collection is performed only with a swab from the buccal mucosa
  • test results are also of a preventive nature - early detection of genetic risks and adequate preventive measures can prevent or at least significantly reduce the consequences of periodontal disease
  • determined markers are related to a number of other systemic diseases: diabetes, rheumatoid arthritis, vascular diseases etc.

How to perform the test?

The test is primarily prepared as an addition to the VariOr® Dento test, but the test can also be performed separately. In this case, the collection is not performed with paper pins, but with a swab from the buccal mucosa.

  • The dentist / periodontist will order a free collection set (VariOr® Dento or VariOr® Gen - unless the doctor is also examining the patient for periodontal pathogens).
  • The dentist / periodontist takes samples according to the instructions included within the set and sends them to the laboratory together with the application form.
  • In the laboratory, the test is performed within 5 working days from sample delivery.
  • The laboratory report is sent to the doctor together with the invoice.

How does the VariOr Dento test work?

The test is based on molecular biological detection of DNA of determined pathogens. The principle of the assay is reverse hybridization of the labeled PCR product with specific membrane-bound probes.
